SYNTHETIC COMPLETE SUPPLEMENT MIXTURE-HIS-LEU-URA - Introduction
SC/-His/-Leu/-Ura Ringer's medium is a common selective medium suitable for the study of yeast genome editing and selection of specific phenotypes. The following is a detailed description of the nature, use, preparation and safety information of the medium:
Nature:
SC/-His/-Leu/-Ura Ringer's medium is a synthetic complete medium containing essential nutrients to meet the growth needs of bacteria or yeast. Their lack of affinity agents (e. g. leucine, histidine and uracil) makes them suitable for yeasts that select leucine, histidine and uracil themselves as nutrient sources.
Use:
SC/-His/-Leu/-Ura medium is commonly used in yeast genome editing and yeast transformation experiments. Due to its lack of affinity agents, only yeasts with leucine, histidine and uracil selection genes were able to grow on this medium.
Preparation Method:
The method for preparing SC/-His/-Leu/-Ura medium is relatively simple. The preparation steps are as follows:
1. Prepare the required chemical reagents, including synthetic complete medium, leucine, histidine and uracil.
2. Prepare according to the protocol for preparing the medium. Usually, the synthetic complete medium is dissolved in an appropriate amount of distilled water, and leucine, histidine and uracil are added in the specified ratio.
3. Stir or shake the solution to evenly mix the nutrients.
4. The solution is sterilized, usually by high-pressure steam sterilization (121 ℃,15-20 minutes) or filtration sterilization (0.22 μm filter membrane).
Safety Information:
The chemical reagents of SC/-His/-Leu/-Ura Ringer's medium have certain safety risks, so appropriate laboratory safety procedures and personal protective measures should be followed during preparation and use. This includes wearing suitable protective eyewear, laboratory coats, gloves and respiratory protection equipment. At the same time, pay attention to the proper disposal of chemical reagent waste and follow local waste disposal regulations.
Please note that when using any medium or reagent, always understand and follow the details provided by the manufacturer, safety data sheet and label instructions.
